Background:

Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Histone H4 are known to be acetylated on Lysine 5, 8, 12, and 16. Acetylations are important for the regulation of histone deposition, transcriptional activation, DNA replication and repair. Hyper-acetylation of the histone tails is shown to neutralize positive charge and weakens histone-DNA and nucleosome-nucleosome interactions that destabilizes chromatin structure and increases the accessibility of DNA to various DNA-binding proteins.
