Background:

The proteasome is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. The proteasome has an ATP-dependent proteolytic activity.

Paraformaldehyde-fixed, paraffin embedded Human testis; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with PSMA5 Monoclonal Antibody, Unconjugated (bsm-51520M) at 1:25 overnight at 4°C, DAB staining.


Lane 1: K562 cell lysates probed with PSMA5 Monoclonal Antibody, Unconjugated (bsm-51520M) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Human liver tissue lysates; Lane 2: Hela cell lysates; Lane 3: HepG2 cell lysates; Lane 4: MCF-7 cell lysates;Lane 5: Neuro-2a cell lysates; Lane 6: PC-12 cell lysates probed with PSMA5 Monoclonal Antibody, Unconjugated (bsm-51520M) at 1:1000 dilution and 4°C overnight incubation.
