Background:

Apurinic/apyrimidinic (AP) sites occur frequently in DNA molecules by spontaneous hydrolysis, by DNA damaging agents or by DNA glycosylases that remove specific abnormal bases. AP sites are pre-mutagenic lesions that can prevent normal DNA replication so the cell contains systems to identify and repair such sites. Class II AP endonucleases cleave the phosphodiester backbone 5' to the AP site. This gene encodes the major AP endonuclease in human cells. Splice variants have been found for this gene; all encode the same protein. [provided by RefSeq, Jul 2008]

Lane 1: A431; Lane 2: HeLa; Lane 3: HepG2; Lane 4: K562; Lane 5: PC-3 cell lysate at 20 µg per lane, probed with bsm-51448M APEX1 (1518CT337.123.86.269.232) Monoclonal Antibody at 1:2000 dilution and 4℃ overnight incubation, followed by secondary antibody incubation for 60min at room temperature.


Paraformaldehyde-fixed, paraffin embedded human stomach tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 15 minutes; Blocking buffer (3% BSA) at room temperature for 30min; Antibody incubation with APEX1 (15C2) Monoclonal Antibody (bsm-51448M) at 1:25 for 1 hour at 37°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.
