Source: This VAMP8 antibody is generated from a mouse immunized with a KLH conjugated synthetic peptide between 2~24 amino acids from the N-terminal region of human VAMP8.

Purification: Purified monoclonal antibody supplied in PBS with 0.09% (W/V) sodium azide. This antibody is purified through a protein G column, followed by dialysis against PBS.

Storage Buffer: 0.01M TBS(pH7.4) with 1% BSA, 0.02% Proclin300 and 50% Glycerol.

Storage: Shipped at 4°C. Store at -20°C for one year. Avoid repeated freeze/thaw cycles.

Background:

SNAREs, soluble N-ethylmaleimide-sensitive factor-attachment protein receptors, are essential proteins for fusion of cellular membranes. SNAREs localized on opposing membranes assemble to form a trans-SNARE complex, an extended, parallel four alpha-helical bundle that drives membrane fusion. VAMP8 is a SNARE involved in autophagy through the direct control of autophagosome membrane fusion with the lysososome membrane via its interaction with the STX17-SNAP29 binary t-SNARE complex (PubMed:23217709, PubMed:25686604). Also required for dense-granule secretion in platelets (PubMed:12130530). Plays also a role in regulated enzyme secretion in pancreatic acinar cells (By similarity). Involved in the abscission of the midbody during cell division, which leads to completely separate daughter cells (By similarity). Involved in the homotypic fusion of early and late endosomes (By similarity).

VALIDATION IMAGES

Lane 1: HeLa; Lane 2: THP-1; Lane 3: A431 cell lysate at 20 µg per lane, probed with bsm-51432M VAMP8 (1414CT354.12.23.93) Monoclonal Antibody at 1:1000 dilution and 4℃ overnight incubation, followed by secondary antibody incubation for 60min at room temperature.


Paraformaldehyde-fixed, paraffin embedded human brain tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 15 minutes; Blocking buffer (3% BSA) at room temperature for 30min; Antibody incubation with VAMP8 (14C3) Monoclonal Antibody (bsm-51432M) at 1:25 for 1 hour at 37°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.


Lane 1: Human THP-1 cell lysates; Lane 2: Human HeLa cell lysates probed with VAMP8 Monoclonal Antibody, Unconjugated (bsm-51432M) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
