Purified His-tagged MGMT protein was used to produced this monoclonal antibody.
Purification:
Purified monoclonal antibody supplied in PBS with 0.09% (W/V) sodium azide. This antibody is purified through a protein G column, eluted with high and low pH buffers and neutralized immediately, followed by dialysis against PBS.
Storage Buffer:
0.01M TBS(pH7.4) with 1% BSA, 0.02% Proclin300 and 50% Glycerol.

Shipped at 4C. Store at -20C for one year. Avoid repeated freeze/thaw cycles.
Background:
Involved in the cellular defense against the biological effects of O6-methylguanine (O6-MeG) in DNA. Repairs alkylated guanine in DNA by stoichiometrically transferring the alkyl group at the O-6 position to a cysteine residue in the enzyme. This is a suicide reaction: the enzyme is irreversibly inactivated.

Lane 1: MCF-7 Cell lysates; probed with MGMT (888CT22.2.4) Monoclonal Antibody, unconjugated (bsm-51234M) at 1:25 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.
Paraformaldehyde-fixed, paraffin embedded human small intestine tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 15 minutes; Blocking buffer (3% BSA) at room temperature for 30min; Antibody incubation with MGMT (888CT22.2.4) Monoclonal Antibody (bsm-51234M) at 1:25 for 1 hour at 37°C, followed by a conjugated secondary antibody for 20 minutes and DAB staining.
