Paraformaldehyde-fixed, paraffin embedded (rat brain); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (beta-Amyloid) Monoclonal Antibody, Unconjugated (bsm-41470M) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Mouse)(sp-0024) instructionsand DAB staining.
Positive sample（left）：Alzheimer's Disease Brain Negative sample（right）：Normal Human Brain Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Incubation with (beta-Amyloid(1-42)) Monoclona Antibody, Unconjugated (bsm-41470M) at 1:1000 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0024) instructionsand DAB staining.
100 ng Aβ1-42 Peptide (bs-0107P) per lane probed with beta-Amyloid(1-42) polyclonal antibody respectively, unconjugated (bsm-41470M) at 1:1000 dilution and 4°C overnight incubation. Followed by corresponding conjugated secondary antibody incubation at r.t. for 60 min.
