DATASHEET

Host: Sheep

Target Protein: Dynamin (Ser778)

Specificity: Specific for endogenous levels of the ~95 kDa dynamin protein phosphorylated at Ser778. Labels the purified protein phosphorylated in vitro by cdk5 but not by PKC. Does not cross react with other purified substrates of cdk5 (e.g. amphiphysin and synapsin). Immunolabeling is completely eliminated by treatment with λ-phosphatase.

Modification Site: Ser778

Clonality: Polyclonal

Isotype: IgG

Entrez Gene: 140694

Swiss Prot: P21575

Source: Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser778 of rat dynamin, conjugated to keyhole limpet hemocyanin (KLH).

Purification: Antigen Affinity purification from Pooled whole antiserum

Storage Buffer: 10 mM HEPES (pH 7.5), 150 mM NaCl, 100 µg per ml BSA and 50% glycerol.

Storage: Storage at -20°C is recommended, as aliquots may be taken without freeze/thawing due to presence of 50% glycerol. Stable for at least 1 year at -20°C.

Background:

Dynamin is a member of a group of nerve terminal proteins called dephosphins that regulate synaptic vesicle endocytosis (Cousin et al., 2001; Graham et al., 2002; Tsuboi et al., 2002). Cyclin dependent protein kinase 5 phosphorylates dynamin at Ser-774 and Ser-778 that are the phosphorylation sites on dynamin phosphorylated in vivo (Tan et al., 2003). Phosphorylation of these sites on dynamin is thought to play a key role in synaptic vesicle trafficking.
