Background:

Lipopolysaccharide is a potent stimulator of monocytes and macrophages, causing secretion of tumor necrosis factor-alpha (TNF-alpha) and other inflammatory mediators. This gene encodes lipopolysaccharide-induced TNF-alpha factor, which is a DNA-binding protein and can mediate the TNF-alpha expression by direct binding to the promoter region of the TNF-alpha gene. The transcription of this gene is induced by tumor suppressor p53 and has been implicated in the p53-induced apoptotic pathway. Mutations in this gene cause Charcot-Marie-Tooth disease type 1C (CMT1C) and may be involved in the carcinogenesis of extramammary Paget's disease (EMPD). Multiple alternatively spliced transcript variants have been found for this gene.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded Human prostate; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with LITAF Polyclonal Antibody, Unconjugated (bs-55125R) at 1:100 overnight at 4°C, DAB staining.


Lane 1: K562 cell lysates; Lane 2: Hela cell lysates; Lane 3: HepG2 cell lysates; Lane 4: BT-474 cell lysates probed with LITAF Polyclonal Antibody, Unconjugated (bs-55125R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Hela cell lysates; Lane 2: LITAF knockout (KO) HeLa cell lysates probed with LITAF Polyclonal Antibody, Unconjugated (bs-55125R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
