Lane 1: HepG2 Cell lysates; Probed with PPAR Gamma (Ser273) Polyclonal Antibody, unconjugated (bs-4888R) at 1:300 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.
Paraformaldehyde-fixed, paraffin embedded Human lung carcinoma; Antigen retrieval by boiling in sodium citrate buffer(pH6) for 15 minutes; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20 minutes; Antibody incubation with PPAR Gamma(ser273) Polyclonal Antibody, Unconjugated (bs-4888R) at 1:200 overnight at 4°C, followed by a conjugated secondary antibody and DAB staining.

An ELISA was performed using a serial dilution of PPAR Gamma (ser273) Polyclonal Antibody (bs-4888R) in antigen-coated wells. The antigen used was a peptide containing the phosphosite of interest. The plot shows the absorbance against the antibody dilution for phosphopeptide antigen and non-phosphorylated peptide antigen.
Lane 1: Human A431 cell lysates; Lane 2: Mouse Adipocyte lysates; Lane 3: Mouse Lung lysates probed with PPAR Gamma Polyclonal Antibody, Unconjugated (bs-4888R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
