Background:

Functions in the early steps of protein synthesis by forming a ternary complex with GTP and initiator tRNA. This complex binds to a 4S ribosomal subunit, followed by mRNA binding to form a 43S preinitiation complex. Junction of the 6S ribosomal subunit to form the 8S initiation complex is preceded by hydrolysis of the GTP bound to eIF-2 and release of an eIF-2-GDP binary complex. In order for eIF-2 to recycle and catalyze another round of initiation, the GDP bound to eIF-2 must exchange with GTP by way of a reaction catalyzed by eIF-2B.

Paraformaldehyde-fixed, paraffin embedded rat lung tissue; Antigen retrieval by boiling in sodium citrate buffer(pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with Rabbit Anti-eIF2 alpha Polyclonal Antibody, Unconjugated (bs-3613R) at 1:500 overnight at 4°C, followed by a conjugated secondary and DAB staining


Paraformaldehyde-fixed, paraffin embedded Rat brain; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with eIF2 alpha Polyclonal Antibody, Unconjugated (bs-3613R) at 1:400 overnight at 4°C, DAB staining.


Lane 1: Human A431 cell lysates; Lane 2: Human Hela cell lysates; Lane 3: Human Jurkat cell lysates probed with eIF2 alpha Polyclonal Antibody, Unconjugated (bs-3613R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
