Background:
Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

Formalin-fixed and paraffin embedded rat brain labeled with Rabbit Anti Phospho-Histone H3 (Ser10) Polyclonal Antibody, Unconjugated (bs-3186R) at 1:200 followed by conjugation to the secondary antibody and DAB staining
MCF 7 cells were fixed with 70% ice-cold methanol overnight at 4℃,permeabilized with 90% ice-cold methanol for 20 min at -20℃, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Histone H3 (Ser11) Polyclonal Antibody at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed.. Control cells (dark blue), secondary only (light blue), Isotype control (orange) and Histone H3 (Ser11) (green).
