Background:

Transcription factor that is involved in embryonic development, establishment of tissue-specific gene expression and regulation of gene expression in differentiated tissues. Is thought to act as a 'pioneer' factor opening the compacted chromatin for other proteins through interactions with nucleosomal core histones and thereby replacing linker histones at target enhancer and/or promoter sites. Binds DNA with the consensus sequence 5'-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3' (By similarity). In embryonic development is required for notochord formation. Involved in the development of multiple endoderm-derived organ systems such as the liver, pancreas and lungs; FOXA1 and FOXA2 seem to have at least in part redundant roles. Originally described as a transcription activator for a number of liver genes such as AFP, albumin, tyrosine aminotransferase, PEPCK, etc. Interacts with the cis-acting regulatory regions of these genes. Involved in glucose homeostasis; regulates the expression of genes important for glucose sensing in pancreatic beta-cells and glucose homeostasis. Involved in regulation of fat metabolism. Binds to fibrinogen beta promoter and is involved in IL6-induced fibrinogen beta transcriptional activation.

VALIDATION IMAGES

Formalin-fixed and paraffin embedded human prostate tissue labeled with Anti-FOXA2/HNF 3beta Polyclonal Antibody, Unconjugated (bs-2358R) at 1:300 followed by conjugation to the secondary antibody and DAB staining.


F9 cells probed with Rabbit Anti-SP-C Polyclonal Antibody (bs-10067R) at 1:100 for 60 minutes at room temperature followed by Goat Anti-Rabbit IgG (H+L) Alexa Fluor 488 Conjugated Secondary Antibody.


Mouse Placenta lysates probed with FOXA2 Polyclonal Antibody, Unconjugated (bs-2358R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:10000 for 60 min at 37˚C.


HepG2 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 90% ice-cold methanol for 20 min at -20℃, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with FOXA2 Polyclonal Antibody(bs-2358R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).


Lane 1: Mouse Liver tissue lysates; Lane 2: Rat Liver tissue lysates probed with FOXA2 Polyclonal Antibody, Unconjugated (bs-2358R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


25 ug total protein per lane of various lysates (see on figure) probed with FOXA2 polyclonal antibody, unconjugated (bs-2358R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
