Background:
Transmembrane glycoprotein of the endoplasmic reticulum that functions as a transcription activator and initiates the unfolded protein response (UPR) during endoplasmic reticulum stress. Cleaved upon ER stress, the N-terminal processed cyclic AMP-dependent transcription factor ATF-6 alpha translocates to the nucleus where it activates transcription of genes involved in the UPR. Binds DNA on the 5'-CCAC[GA]-3'half of the ER stress response element (ERSE) (5'-CCAAT-N(9)-CCAC[GA]-3') and of ERSE II (5'-ATTGG-N-CCACG-3'). Binding to ERSE requires binding of NF-Y to ERSE. Could also be involved in activation of transcription by the serum response factor. May play a role in foveal development and cone function in the retina.

Lane 1: Rat cerebrum lysates; Lane 2: MCF-7 lysates; Lane 3: K562 lysates; probed with ATF6 Polyclonal Antibody, unconjugated (bs-23094R) at 1:300 overnight at 4°C followed by a conjugated secondary antibody for 60 minutes at 37°C.
Paraformaldehyde-fixed, paraffin embedded rat brain; Antigen retrieval by boiling in sodium citrate buffer (pH6) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20min; Antibody incubation with ATF6 Polyclonal Antibody, Unconjugated (bs-23094R) at 1:200 overnight at 4°C, followed by a conjugated secondary and DAB staining.
Lane 1: Human HUVEC cell Lysates; Lane 2: Human Raji cell Lysates; Lane 3: Human Jurkat cell Lysates; Lane 4: Human Hela cell Lysates. Probed with ATF6 polyclonal Antibody, unconjugated (bs-23094R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
