Background:

The enzyme apparently serves as a quinone reductase in connection with conjugation reactions of hydroquinons involved in detoxification pathways as well as in biosynthetic processes such as the vitamin K-dependent gamma-carboxylation of glutamate residues in prothrombin synthesis.

VALIDATION IMAGES

Formalin-fixed and paraffin embedded mouse colon carcinoma labeled with Anti-NQO1 Polyclonal Antibody, Unconjugated (bs-2184R) at 1:200, followed by conjugation to the secondary antibody and DAB staining


L1 rat liver lysates L2 rat brain lysates probed with Anti NQO1 Polyclonal Antibody, Unconjugated (bs-2184R) at 1:200 overnight at 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000 for 90 min at RT. Predicted band 30kD. Observed band size:30kD.\n


Lane 1: A549 cell lysates probed with NQO1 Polyclonal Antibody, Unconjugated (bs-2184R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Human Hela cell lysates; Lane 2: Human HepG2 cell lysates; Lane 3: Human MCF-7 cell lysates; Lane 4: Human Du145 cell lysates probed with NQO1 Polyclonal Antibody, Unconjugated (bs-2184R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Paraformaldehyde-fixed, paraffin embedded (rat stomach); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (NQO1) Polyclonal Antibody, Unconjugated (bs-2184R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.


4% Paraformaldehyde-fixed HepG2 (H) cell; Triton X-100 at r.t. for 20 min; Antibody incubation with (NQO1) polyclonal Antibody, unconjugated (bs-2184R) 1:100, 90 min at 37°C; followed by conjugated Goat Anti-Rabbit IgG antibody (green, bs-40295G-FITC) at 37°C for 90 min, DAPI (blue, C02-04002) was used to stain the cell nuclei. PBS instead of the primary antibody was used as the blank control.


25 ug total protein per lane of various lysates (see on figure) probed with NQO1 polyclonal antibody, unconjugated (bs-2184R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
