Background:

Tau proteins are important Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization. Tau proteins subcellular located in the axons of neurons, in the cytoso l and in association with plasma membrane components. It expressed in neurons. PNS-tau is expressed in the peripheral nervous system while the others are expressed in the central nervous system.

VALIDATION IMAGES

Human U251 lysates probed with Tau Polyclonal Antibody, Unconjugated (bs-20445R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:10000 for 60 min at 37˚C.


Paraformaldehyde-fixed, paraffin embedded Mouse cerebellum tissue; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with Tau Polyclonal Antibody, Unconjugated (bs-20445R) at 1:400 overnight at 4°C, followed secondary and DAB staining.

Human MCF-7 Cells fixed with 70% methanol (Overnight at 4℃) and then permeabilized with 90% ice-cold methanol for 30 min on ice. Cells were then probed with rabbit anti-Tau Polyclonal Antibody, Unconjugated (bs-20445R) (green) at 1:33 for 30 minutes followed by a FITC conjugated secondary antibody compared to unstained cells (dark blue), secondary only (light blue), and isotype control (orange).
