Background:

Cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied transferrin receptor into specialized endosomes. Endosomal acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. Transferrin receptor is necessary for development of erythrocytes and the nervous system (By similarity). A second ligand, the heditary hemochromatosis protein HFE, competes for binding with transferrin for an overlapping C-terminal binding site.

VALIDATION IMAGES

Mouse placenta lysates probed with Rabbit Anti-Transferrin receptor Polyclonal Antibody, Unconjugated (bs-0988R) at 1:300 overnight at 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:500 for 90 min at RT


Raji cells probed with \tTransferrin receptor Polyclonal Antibody, Unconjugated (bs-0988R) at 1:100 for 30 minutes followed by incubation with a conjugated secondary (PE Conjugated) (green) for 30 minutes compared to control cells (blue), secondary only (light blue) and isotype control (orange).


K562 cells were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Transferrin receptor Polyclonal Antibody(bs-0988R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).


Paraformaldehyde-fixed, paraffin embedded (human liver); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (TFRC ) Polyclonal Antibody, Unconjugated (bs-0988R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
