Background:
Expression of the POU-domain transcription factor Octamer-4 (Oct-4) is widely regarded as a hallmark of pluripotent stem cells. The relationship of Oct-4 to pluripotent stem cells is indicated by its tightly restricted expression to undifferentiated pluripotent stem cells. Upon differentiation to somatic lineages, the expression of Oct-4 disappears rapidly. Unlike the majority of pluripotent stem cell markers, the biological role of Oct-4 has been well characterized. Studies performed in mice point to the critical role of Oct-4 in the establishment and/or maintenance of pluripotent stem cells in an uncommitted state.

VALIDATION IMAGES
L1 rat brain, L2 , human colon carcinoma lysates probed (bs-0830R) at 1:200 in 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000 90min in RT. Predicted and observed band size: 41kDa.
Formalin-fixed and paraffin embedded Human osteosarcoma labeled with Rabbit Anti-Oct-4/POU5F1 Polyclonal Antibody, Unconjugated (bs-0830R) 1:300 followed by conjugation to the secondary antibody and DAB staining.
Lane 1: Mouse Cerebrum lysates; Lane 2: Mouse Heart lysates; Lane 3: Mouse Cerebellum lysates; Lane 4: Mouse Testis lysates; Lane 5: Rat Cerebrum lysates; Lane 6: Rat Heart lysates; Lane 7: Rat Cerebellum lysates probed with Oct4 Polyclonal Antibody, Unconjugated (bs-0830R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
