Background:

This gene encodes the pro-alpha1 chains of type I collagen whose triple helix comprises two alpha1 chains and one alpha2 chain. Type I is a fibril-forming collagen found in most connective tissues and is abundant in bone, cornea, dermis and tendon. Mutations in this gene are associated with osteogenesis imperfecta types I-IV, Ehlers-Danlos syndrome type VIIA, Ehlers-Danlos syndrome Classical type, Caffey Disease and idiopathic osteoporosis. Reciprocal translocations between chromosomes 17 and 22, where this gene and the gene for platelet-derived growth factor beta are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans, resulting from unregulated expression of the growth factor. Two transcripts, resulting from the use of alternate polyadenylation signals, have been identified for this gene. Type I collagen levels have been linked to bone growth and formation. Levels of the C-terminal propeptide are indicative of collagen production in vivo. Elevated levels of C-terminal propeptide have been shown in diseases associated with high levels of bone turnover, including Pagets disease, hyperthyroidism, primary hyperparathyroidism and renal osteodystrophy. Slightly elevated levels of C-terminal propeptide have also been documented in some women in early menopause.

Formalin-fixed and paraffin-embedded rabbit meniscus tissue labeled with Anti-Collagen I Polyclonal Antibody, Unconjugated (bs-0578R) 1:300 followed by conjugation to the secondary PE antibody and DAB staining


Paraformaldehyde-fixed, paraffin-embedded Human Lung Cancer tissue; Antigen retrieval by boiling in sodium citrate buffer (0.01M, pH6) for 15 minutes; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (normal goat serum) at 37°C for 20 minutes; Antibody incubation with Pro-Collagen 1 Polyclonal Antibody, Unconjugated (bs-0578R) at 1:200 overnight at 4°C, followed by a conjugated secondary antibody, DAB and counterstaining.


Lane 1: Mouse Placenta tissue lysates; Lane 2: Human U-2OS cell lysates probed with Collagen I Polyclonal Antibody, Unconjugated (bs-0578) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at room temperature for 60 min.


Blank control:Hela. Primary Antibody (green line): Rabbit Anti-Collagen I antibody (bs-0578R) Dilution: 1ug/Test; Protocol Hela cells were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Collagen I Polyclonal Antibody(bs-0578R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
