Background:

Participates in various redox reactions through the reversible oxidation of its active center dithiol to a disulfide and catalyzes dithiol-disulfide exchange reactions. Plays a role in the reversible S-nitrosylation of cysteine residues in target proteins, and thereby contributes to the response to intracellular nitric oxide. Nitrosylates the active site Cys of CASP3 in response to nitric oxide (NO), and thereby inhibits caspase-3 activity. Induces the FOS/JUN AP-1 DNA-binding activity in ionizing radiation (IR) cells through its oxidation/reduction status and stimulates AP-1 transcriptional activity. ADF augments the expression of the interleukin-2 receptor TAC (IL2R/P55).

Paraformaldehyde-fixed, paraffin embedded human cervical carcinoma; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with Thioredoxin Polyclonal Antibody, Unconjugated (bs-0458R) at 1:400 overnight at 4°C, followed by a conjugated secondary for 20 minutes and DAB staining.


Lane 1: Trx-Tagged fusion protein (full length) Overexpression E.coli lysates (Cat#: bs-33018P) probed with Thioredoxin/Trx Polyclonal Antibody, Unconjugated (bs-0458R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Hela cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (Thioredoxin) polyclonal Antibody, Unconjugated (bs-0458R) 1:100, 90 minutes at 37°C; followed by a conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.
