Background:

Ubiquitinous metalloproteinase that is involved in diverse functions such as remodeling of the vasculature, angiogenesis, tissue repair, tumor invasion, inflammation, and atherosclerotic plaque rupture. As well as degrading extracellular matrix proteins, can also act on several nonmatrix proteins such as big endothelial 1 and beta-type CGRP promoting vasoconstriction. Also cleaves KISS at a Gly-|-Leu bond. Appears to have a role in myocardial cell death pathways. Contributes to myocardial oxidative stress by regulating the activity of GSK3beta. Cleaves GSK3beta in vitro. Involved in the formation of the fibrovascular tissues in association with MMP14. PEX, the C-terminal non-catalytic fragment of MMP2, posseses anti-angiogenic and anti-tumor properties and inhibits cell migration and cell adhesion to FGF2 and vitronectin. Ligand for integrinv/beta3 on the surface of blood vessels. Isoform 2: Mediates the proteolysis of CHUK/IKKA and initiates a primary innate immune response by inducing mitochondrial-nuclear stress signaling with activation of the pro-inflammatory NF-kappaB, NFAT and IRF transcriptional pathways.

Formalin-fixed and paraffin embedded rat carotid artery Anti-MMP-2 Polyclonal Antibody (bs-0412R), Unconjugated at 1:200, followed by conjugation to the secondary antibody and DAB staining.


Paraformaldehyde-fixed, paraffin embedded rat brain; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with MMP2 Polyclonal Antibody, Unconjugated bs-0412R at 1:500 overnight at 4°C, followed by a conjugated secondary for 20 minutes and DAB staining.

HT1080 lysates probed with MMP-2 Polyclonal Antibody, Unconjugated (bs-0412R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:10000 for 60 min at 37˚C.


Lane 1: Mouse Ovary lysates; Lane 2: Mouse Heart lysates; Lane 3: Mouse Stomach lysates probed with MMP2 Polyclonal Antibody, Unconjugated (bs-0412R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Human SH-SY5Y cell lysates; Lane 2: Human U2os cell lysates; Lane 3: Human U251 cell lysates; Lane 4: Human Siha cell lysates probed with MMP-2 Polyclonal Antibody, Unconjugated (bs-0412R) at 1:1000 dilution and 4°C overnight incubation.
