Background:

Integrin alpha-V/beta-3 is a receptor for cytotactin, fibronectin, laminin, matrix metalloproteinase-2, osteopontin, osteomodulin, prothrombin, thrombospondin, vitronectin and von Willebrand factor. Integrin alpha-IIb/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. Integrins alpha-IIb/beta-3 and alpha-V/beta-3 recognize the sequence R-G-D in a wide array of ligands. Integrin alpha-IIb/beta-3 recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain. Following activation integrin alpha-IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation which physically plugs ruptured endothelial surface. Fibrinogen binding enhances SELP expression in activated platelets (By similarity). In case of HIV-1 infection, the interaction with extracellular viral Tat protein seems to enhance angiogenesis in Kaposi's sarcoma lesions.

VALIDATION IMAGES

Formalin-fixed and paraffin embedded human rectal carcinoma labeled with Anti-Integrin beta 3/CD61 Polyclonal Antibody, Unconjugated (bs-0342R) at 1:200, followed by conjugation to the secondary antibody and DAB staining


Human HL-60 cells probed with Integrin beta 3 Polyclonal Antibody, Unconjugated (bs-0342R) (green) at 1:100 for 30 minutes followed by a FITC conjugated secondary antibody compared to unstained cells (blue), secondary only (light blue), and isotype control (orange).


HL-60 (Positive) and A431 (Negative control) cells (black) were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Integrin beta 3 Antibody (bs-0342R) at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS, followed by secondary antibody(blue) incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).


Lane 1: Mouse Bone Lysates; Lane 2: Mouse Kidney Lysates. Probed with Integrin beta 3polyclonal Antibody, unconjugated (bs-0342R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
