Background:

Receptor for TNFSF6/FASLG. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both (By similarity).

Formalin-fixed and paraffin embedded rat colon tissue labeled with Anti-Fas Polyclonal Antibody, Unconjugated (bs-0215R) at 1:200 followed by conjugation to the secondary antibody and DAB staining

Lane 1: Mouse Thymus tissue lysates; Lane 2: Rat Thymus tissue lysates; Lane 3: Human HepG2 cell Lysates; Lane 4: Human Raji cell Lysates; Lane 5: Human HT1080 cell Lysates; Lane 6: Human A431 cell Lysates probed with CD95/FAS Polyclonal Antibody, Unconjugated (bs-0215R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at room temperature for 60 min.

Raji cells were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with CD95/FAS Polyclonal Antibody(bs-0215R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
