Background:

Big Endothelin-1 is precursor of endothelin 1. Big endothelin-1 is cleaved to yield endothelin-1 via the activity of an endothelin-converting enzyme (ECE). Big Endothelin-1 can be hydrolyzed by chymase to generate endothelin 1 (1-21) in vitro. Endothelins are endothelium-derived vasoconstrictor peptides.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded Human Stomach; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with ET-1 Polyclonal Antibody, Unconjugated (bs-0202R) at 1:200 overnight at 4°C, DAB staining.


Lane 1: Mouse Testis tissue lysates; Lane 2: Rat Testis tissue lysates probed with Big endothelin-1 Polyclonal Antibody, Unconjugated (bs-0202R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at room temperature for 60 min.
