Background:

In cooperation with other chaperones, Hsp7s stabilize preexistent proteins against aggregation and mediate the folding of newly translated polypeptides in the cytosol as well as within organelles. These chaperones participate in all these processes through their ability to recognize nonnative conformations of other proteins. They bind extended peptide segments with a net hydrophobic character exposed by polypeptides during translation and membrane translocation, or following stress-induced damage. In case of rotavirus A infection, serves as a post-attachment receptor for the virus to facilitate entry into the cell.

Generously provided by Merlin Levine from National Institutes of Health as part of the Bioss Discovery Program. Mouse brain labeled with Anti-HSP70 Polyclonal Antibody, Unconjugated (bs-0126R) at 1:100 overnight. The secondary antibody was AlexaFluor 555 goat anti mouse IgG1 used at 1:250 for one hour.


Mouse bladder lysates probed with HSP70 Polyclonal Antibody, Unconjugated (bs-0126R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:10000 for 60 min at 37˚C.


Hela cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (HSP70) polyclonal Antibody, Unconjugated (bs-0126R) 1:100, 90 minutes at 37°C; followed by a conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.
