Background:

Multifunctional protein that controls proliferation, differentiation and other functions in many cell types. Many cells synthesize TGFB1 and have specific receptors for it. It positively and negatively regulates many other growth factors. It plays an important role in bone remodeling as it is a potent stimulator of osteoblastic bone formation, causing chemotaxis, proliferation and differentiation in committed osteoblasts.

VALIDATION IMAGES

Formalin-fixed and paraffin embedded rat renal tissue labeled with Anti-TGF-beta-1 Polyclonal Antibody, Unconjugated(bs-0086R) 1:200 followed by conjugation to the secondary antibody and DAB staining


Paraformaldehyde-fixed, paraffin embedded Rat Lung; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with TGF beta 1 Polyclonal Antibody, Unconjugated (bs-0086R) at 1:500 overnight at 4°C, DAB staining.


Lane 1: Mouse Embryo; Lane 2: Rat Cerebrum; Probed with TGF beta 1 Polyclonal Antibody, Unconjugated (bs-0086R) at 1:1000 dilution, overnight at 4˚C. Followed by secondary antibody incubation at 1:10000 for 60 min at 37˚C.


Lane 1: Isolated Transforming growth factor beta-1; Probed with TGF beta 1 Polyclonal Antibody, Unconjugated (bs-0086R) at 1:300 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:10000 for 60 min at 37˚C.


Lane 1: Mouse Ovary; Probed with TGF beta 1 Polyclonal Antibody, Unconjugated (bs-0086R) at 1:1000 dilution, overnight at 4˚C. Followed by secondary antibody incubation at 1:10000 for 60 min at 37˚C.


Lane 1: Mouse Liver lysates; Lane 2: Rat Liver lysates probed with TGF beta 1 Polyclonal Antibody, Unconjugated (bs-0086R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


Lane 1: Mouse Stomach lysates; Lane 2: Mouse Liver lysates; Lane 3: Mouse Placenta lysates; Lane 4: Mouse NIH/3T3 cell lysates; Lane 5: Rat Liver lysates probed with TGF beta 1 Polyclonal Antibody, Unconjugated (bs-0086R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.


MG63 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1%PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with TGF beta 1 Polyclonal Antibody(bs-0086R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).


25 ug total protein per lane of various lysates (see on figure) probed with TGF Beta 1 polyclonal antibody, unconjugated (bs-0086R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
